dualguide - progress

  • goal: demonstrate the importance of barcoding by showing that relying solely on tumor burden results in loss of information
  • volcano plots (fig1_volcano_plots.pdf)
    • x-axis: log2(original value), y-axis: -log10(p value)
    • 90th percentile, ln_mean, n_of_tumors, tumor burden
    • results
      • more negative effects with number of tumors
      • possible correlation between 90th and lnMean and tumor burden
    • note
      • remember we discussed that the titer was not calculated as usual, if exact numbers are needed, i will verify them
  • "4type plots" (fig2_4type_plots.pdf)
    • two growth measurements, a pair (ie. Apc_Lkb1 and we calculate the effect from Apc and from Lkb1 as well, so more points on the plot)
      • 4 types:
        • not significant with either growth measurements
        • significant with both growth measurements
        • significant with only the first gm
        • significant with only the second gm
    • our plots: one growth measurement is always the tumor burden, so we have three plots:
      • tb vs 90th, tb vs lnmean, tb vs number of tumors
      • significant results
        • 90th << tumor burden
        • lnmean < tumor burden
        • number of tumors >> tumor burden
  • "arrow figures" (fig3_arrow_figures.pdf)
    • for each pair we have a figure for each growth measurements
    • y-axis: growth measurement value
    • x-axis: inert - A - AB - B - inert
    • arrows
      • color: positive or negative effect
      • solid-dashed: significant or not
    • different results with each growth measurements
